RE SEARCH The new inspections 8 In order to isolate the released DNA, the well plates enriched with the buffers are placed in another advanced piece of laboratory equipment. The technicians now add magnetic particles to the plant juice, which also pick up DNA. This special machine washes out the DNA, after which it is released from the magnetic particles. This process lasts approximately 40 minutes. We now have purified DNA that can be analysed with the PCR machine. 10 The following step involves pipetting precisely-measured quantities of fluid from the well plates (necessary for DNA testing with the PCR equipment) and transferring them to other well plates. For this purpose, the NAK uses an advanced pipetting robot. This robot saves many hours of manual work. 9 As soon as the well plates with the exact amount of fluid have been filled, they go to a 96-well pipetting robot, which adds special buffer fluids to each well. Here, the various buffers are indicated with the letters B, C and D. So an exact amount of each buffer is added to the sample fluid. The buffer fluids are necessary to single out the DNA. They break up the cells in the juice so that the DNA can be released. Meanwhile, the NAK now has the use of a series of machines that, with the help of PCR (Polymerase Chain Reaction), are able to analyse very small amounts of specific DNA. This means that it is finally possible to show whether DNA of a certain virus or bacterium is present in a sample. In order to be able to do this fast, the laboratory in Emmeloord uses Realtime-PCR. Put simply, the machine copies the specific DNA of any present virus and/or bacterium (Erwinia). The exponential increase of the specific DNA creates fluorescent light. Each wave length can be represented by a colour that, in turn, is specific for each type of virus or bacterium. If a specific virus or bacterium is present, it is shown on the computer screen in various colours in a graph. The computer then converts the result of the analysis into a final result.
